When using dowel joinery, it's not uncommon that one of the holes fits the dowel a little loosely. The hole might be too large or misshapen for a proper fit.
Here's an easy fix: Mix some fine wood chips and sawdust with wood glue to make a paste. Roll the dowel in the mixture and insert it into the hole. The sawdust acts as a filler for any gaps around the dowel.
